Regulation of MHC Class I by Viruses
نویسندگان
چکیده
Originally recognized for their role in triggering T cell responses that caused the rejection of transplanted tissue, it is now known that MHC-encoded molecules are involved in immunesurveillance and antigen presentation to T lymphocytes. These MHC molecules act as “signposts” that displays fragmented pieces of an antigen (viral proteins in virus-infected cells or mutated proteins in tumor cells) on the host cell surface. Because viruses can infect virtually all nucleated cells, class I molecules are constitutively expressed on almost all nucleated cells. The association of antigenic peptides and MHC molecules is a saturable interaction and once formed, persist for a sufficiently long time to be recognized by the few T cells specific for the antigen as they circulate through. The outcome of TC cells-mediated killing of the virus-infected cells is usually via apoptosis.

Human Papillomavirus and Cancer - Immunological Consequences of MHC Class I Down-Regulation
Few studies have been conducted on the causative factors associated with the development of cancer. Infection by high risk human papillomaviruses (HPVs) have been implicated as causative agents in a variety of cancers. HPV is capable of evading immune system and establishing persistent infections. Prolonged infection and lesion maintenance are associated with higher risk of neoplastic progressi...
متن کاملSUSCEPTIBILITY OF HUMAN WM MELANOMA CELL LINES TO NK AND LAK CYTOTOXICITY AND THEIR RELEVANCE TO THE LEVEL OF MHC CLASS I AND ICAM-l ANTIGEN EXPRESSION
The effect of natural killer (NK) cells and lymphokine activated killer ( LAK) cells was studied on a group of human melanoma cell lines. Peripheral blood from healthy volunteers was utilized as a fresh source of natural killer cells and rhI L-2 for producing LAK cells. The cytotoxicity of effector cells was quantified using a 4 hour SI determining the density of antigen expression on tumor...
متن کاملStructural basis for recognition of MHC and MHC-like ligands by natural killer cell receptors.
Natural killer (NK) cell function is regulated by NK cell receptors that bind classical MHC class I molecules or their structural relatives. The latter group includes self-ligands (MICA, RAE-I, H-60), as well as ligands encoded by viruses (UL18, m155, m157). Two distinct families of NK receptors have been identified: the immunoglobulin-like family (KIRs, LIRs) and the C-type lectin-like family ...
متن کاملThe SH3 domain-binding surface and an acidic motif in HIV-1 Nef regulate trafficking of class I MHC complexes.
Nef, a regulatory protein of human and simian immunodeficiency viruses, downregulates cell surface expression of both class I MHC and CD4 molecules in T cells by accelerating their endocytosis. Fibroblasts were used to study alterations in the traffic of class I MHC complexes induced by Nef. We found that Nef downregulates class I MHC complexes by a novel mechanism involving the accumulation of...
متن کاملVirus Encoded MHC-Like Decoys Diversify the Inhibitory KIR Repertoire
Natural killer (NK) cells are circulating lymphocytes that play an important role in the control of viral infections and tumors. Their functions are regulated by several activating and inhibitory receptors. A subset of these receptors in human NK cells are the killer immunoglobulin-like receptors (KIRs), which interact with the highly polymorphic MHC class I molecules. One important function of...

Murine cytomegalovirus (MCMV) infection of C57BL/6 (H-2b) mice prevents priming of antigen-specific helper and cytotoxic T lymphocytes (CTL) (J. S. Slater, W. S. Futch, V. J. Cavanaugh, and A. E. Campbell, Virology 185:132-139, 1991). In vitro, MCMV infection alters presentation of antigens to antigen-specific, MHC class I-restricted CTL (A. E. Campbell, J. S. Slater, V. J. Cavanaugh, and R.
